Mechanism

ACTH stimulates the synthesis and release of adrenocortical hormones by increasing de novo biosynthesis (steroid hormones are hydrophobic and thus cannot be stored). ACTH binding to MC2R activates the Gs–adenylyl cyclase–cyclic AMP–PKA pathway. Cyclic AMP is the second messenger for most effects of ACTH on steroidogenesis.
