Mechanism
Ixabepilone binds to a β-tubulin pocket located on the luminal side of microtubules (the taxane site) and can trigger microtubule nucleation at multiple sites away from the centriole. This dysfunctional microtubule stabilization triggers cell-cycle arrest at the G2-M interface and apoptosis.
Indications
Metastatic breast cancer (resistant to or pretreated with anthracyclines and resistant to taxanes)Metastatic breast cancer (monotherapy in patients who have previously progressed through treatment with anthracyclines, taxanes, and capecitabine)
Dosing
- Adult
- Approximately 40 mg/m2 administered as monotherapy or in combination with capecitabine over 3 h every 3 weeks. Patients should be premedicated with both an H1 and an H2 antagonist before receiving ixabepilone to minimize hypersensitivity reactions.…
