Mechanism

Gastrokinetic and LES tonic action due to 5-HT4 agonistic (major) and 5-HT3 antagonistic (minor) action in the myenteric plexus.

Indications

nonulcer dyspepsiadiabetic gastroparesisgastroesophageal reflux disease (GERD, as adjuvant to H2 blockers/PPIs)some cases of chronic constipation

Dosing

Adult
5 mg TDS
Geriatric
2.5 mg TDS

Contraindications

  • concomitant use with CYP3A4 inhibitors (erythromycin, other CYP3A4 inhibitors)
