Mechanism

Trifluridine is a fluorinated pyrimidine nucleoside that inhibits replication of herpesviruses, including acyclovir-resistant strains, and also inhibits cellular DNA synthesis at relatively low concentrations. Trifluridine monophosphate irreversibly inhibits thymidylate synthase, and trifluridine triphosphate is a competitive inhibitor of thymidine triphosphate incorporation into DNA. Trifluridine is incorporated into viral and cellular DNA.

Dosing

Adult
Instillation of one drop into the affected eye(s) every 2 h while awake for the first 2 weeks and then four times daily for an additional 2 weeks.
