Mechanism

Vinblastine is a vinca alkaloid that binds to β-tubulin at a distinct site (the vinca domain), inhibiting tubulin polymerization into microtubules and causing dissolution of the mitotic spindle, arresting cells in metaphase. Its dose-limiting toxicity is myelosuppression (particularly leukopenia), in contrast to vincristine where neurotoxicity predominates — a difference attributed to differential accumulation in bone marrow versus neuronal tissue. Vinblastine is a key component of ABVD chemotherapy for Hodgkin lymphoma.
