Mechanism
A PTHrP(1–34) analogue that increases new bone formation. It permits greater bone formation with lower stimulation of bone resorption compared to teriparatide.
Indications
postmenopausal osteoporosis in women at high risk for fracturessevere osteoporosis in patients at a high risk for fracture
Dosing
- Adult
- Starting dose of 80 μg administered by subcutaneous injection into the periumbilical region of the abdomen. The site of administration should be rotated, but the time of injection should be the same each day.

Contraindications
- Paget disease of bone
- unexplained elevations of alkaline phosphatase
- open epiphyses
- prior radiation therapy involving the skeleton
- patients at increased baseline risk for osteosarcoma
Side effects
Serious
- hypercalcemia (lower incidence than teriparatide)
- orthostatic hypotension (shortly after injection)
- osteosarcoma
