Mechanism
Evinacumab-dgnb is a recombinant human monoclonal antibody that binds to and inhibits angiopoietin-like protein 3 (ANGPTL3). ANGPTL3 normally inhibits lipoprotein lipase (LPL) and endothelial lipase (EL). By inhibiting ANGPTL3, evinacumab-dgnb rescues LPL and EL activities, leading to reduced LDL-C (independent of LDL receptor), triglycerides, and HDL-C, and promotes VLDL processing and clearance.
Indications
Adjunct to lipid-lowering medications and diet for patients with HoFH
Dosing
- Adult
- 15 mg/kg administered by intravenous infusion over 60 min every 4 weeks
Pharmacokinetics
Onset
Median time for serum concentrations to decrease below the lower limit of quantitation is 19 weeks after the last steady-state dose.
Excretion
Degraded into small peptides and amino acids via catabolic pathways like endogenous immunoglobulin G (IgG)
