Mechanism

Flutamide is an androgen receptor antagonist that inhibits the binding of androgens to the androgen receptor. When used in conjunction with GnRH analogs, it blocks the action of adrenal androgens, which are not inhibited by GnRH analogs. Increased LH secretion can stimulate higher serum testosterone concentrations when used alone, limiting efficacy.

Indications

Advanced prostate cancerMetastatic prostate cancer refractory to gonadorelin analogue therapy (monotherapy)Metastatic prostate cancer (in conjunction with a GnRH analog)Hirsutism in women (off-label)Familial GnRH-independent sexual precocity with premature Leydig cell maturation (in combination with testolactone)hirsutismAll forms of advanced prostate cancer (used infrequently due to unfavorable toxicity profile)metastatic carcinoma prostate (as part of combined androgen blockade therapy)Metastatic prostate cancer
