Mechanism
GnRH is a hypothalamic decapeptide that stimulates pituitary gonadotropin production (LH and FSH) when secreted pulsatilely. It signals through a unique GPCR on gonadotropes, leading to increased LH and FSH secretion and expression of glycoprotein hormone subunit genes.
Indications
Induction of sexual maturation (if fertility is the goal)
Dosing
- Adult
- Intermittent release is crucial for proper synthesis and release of gonadotropins; continuous administration leads to desensitization and downregulation of GnRH receptors.
Pharmacokinetics
Duration
shorter half-life than synthetic analogues
