Mechanism

GnRH is a hypothalamic decapeptide that stimulates pituitary gonadotropin production (LH and FSH) when secreted pulsatilely. It signals through a unique GPCR on gonadotropes, leading to increased LH and FSH secretion and expression of glycoprotein hormone subunit genes.

Indications

Induction of sexual maturation (if fertility is the goal)

Dosing

Adult
Intermittent release is crucial for proper synthesis and release of gonadotropins; continuous administration leads to desensitization and downregulation of GnRH receptors.

Pharmacokinetics

Duration
shorter half-life than synthetic analogues
