Mechanism
Inclisiran is a small interfering ribonucleic acid (siRNA) therapeutic that targets PCSK9 mRNA. By blocking PCSK9 protein synthesis, it prevents the production of PCSK9, leading to reduced degradation of LDL receptors and increased liver clearance of LDL from the circulation, thereby lowering serum LDL-C levels.
Indications
Adjunct to diet and maximally tolerated statin therapy for treatment of adults with HeFH who require additional lowering of LDL-CAdjunct to diet and maximally tolerated statin therapy for treatment of adults with clinical ASCVD who require additional lowering of LDL-C
Dosing
- Adult
- 284 mg in 1.5 mL administered as a single subcutaneous injection initially, then after 3 months, and every 6 months thereafter.
- Renal adjustment
- No dosage adjustments required in patients with mild to severe renal impairment.
- Hepatic adjustment
- No dosage adjustments required in patients with mild to moderate liver impairment.
