Mechanism

Sulfonamides are transformed into hydroxylamines that interact with cellular proteins, generating haptens that can elicit autoimmune responses.

Pharmacokinetics

Metabolism
Sulfonamides undergo acetylation.
Excretion
Sulfonamides are excreted directly in the colon.

Contraindications

  • Slow acetylator phenotype (predisposed to autoimmune responses and hypersensitivity reactions)
  • Potentially nephrotoxic (due to crystalluria)

Side effects

Common
chronic and low grade phototoxic photosensitivityphotoallergic photosensitivity
Serious
  • Idiosyncratic hypersensitivity reactions
  • haemolysis (in G-6PD deficient individuals)
