Mechanism

Telotristat ethyl reduces diarrhea by inhibiting tryptophan hydroxylase, the rate-limiting enzyme of 5HT biosynthesis. Reduced 5HT secretion decreases fluid secretion and motility in the GI tract.

Indications

Diarrhea associated with carcinoid tumors (in combination with somatostatin analogue therapy)

Dosing

Adult
250 mg three times per day (in combination with somatostatin analogue therapy for diarrhea associated with carcinoid tumors).

Pharmacokinetics

Peak effect
1 to 3 h (for active agent telotristat)
Half-life
5 h (for active agent telotristat)
