Mechanism

Basiliximab is a monoclonal antibody that acts as an interleukin-2 receptor antagonist, preventing T-lymphocyte proliferation. It targets the CD25 subunit of the interleukin-2 receptor (IL-2R) on T-cells, thereby limiting clonal expansion and promoting immune tolerance.

Indications

Prophylaxis of acute rejection in allogeneic renal transplantation (used in combination with ciclosporin and corticosteroid-containing immunosuppression regimens)Prophylaxis of graft rejection in cardiac transplantation (off-label, increased risk of serious cardiac side-effects)prophylaxis of acute organ transplant rejectioninduction therapy in solid-organ transplantation

Dosing

Adult
Initially 20 mg, administered within 2 hours before transplant surgery, followed by 20 mg after 4 days. Dose to be administered after surgery. Withhold second dose if severe hypersensitivity or graft loss occurs. Administered by intravenous injection or intravenous infusion.

Pharmacokinetics

Half-life
7 days

Contraindications

  • Severe hypersensitivity (subsequent doses)
