Mechanism

C1-esterase inhibitor (C1-INH) is a plasma protein that regulates the complement and contact systems. Hereditary angioedema (HAE) results from a deficiency or dysfunction of endogenous C1-INH, leading to uncontrolled activation of these systems and increased bradykinin production, which mediates angioedema attacks. Cinryze provides exogenous C1-esterase inhibitor, thereby replacing the deficient protein to regulate these pathways and prevent or treat HAE attacks.

Acute attacks of hereditary angioedema (under expert supervision)Short-term prophylaxis of hereditary angioedema before dental, medical, or surgical procedures (under expert supervision)Long-term prophylaxis of severe, recurrent attacks of hereditary angioedema where acute treatment is inadequate, or when oral prophylaxis is inadequate or not tolerated (under expert supervision)

Dosing

Adult
Acute attacks of hereditary angioedema: 1000 units for 1 dose by slow intravenous injection, dose may be repeated if necessary after 60 minutes (or sooner for patients experiencing laryngeal attacks or if treatment initiation is delayed). Short-term prophylaxis: 1000 units for 1 dose by slow intravenous injection, to be administered up to 24 hours before procedure.…
