Mechanism

Denosumab is a human monoclonal antibody that inhibits osteoclast formation, function, and survival, thereby decreasing bone resorption.

Indications

Osteoporosis in postmenopausal women and in men at increased risk of fracturesBone loss associated with hormone ablation in men with prostate cancer at increased risk of fracturesAs an alternative for osteoporosis in men intolerant of oral bisphosphonates or when bisphosphonates are contra-indicatedAs an alternative for osteoporosis in women intolerant of oral bisphosphonates or when bisphosphonates are contra-indicatedtreatment and prevention of osteoporosistreatment to increase bone mass in adults at high risk for fracture receiving cancer therapyPostmenopausal osteoporosis (when no other drug is appropriate)For patients whose bone health is compromised by breast cancer therapyAdministered regularly for metastatic bone disease to reduce skeletal morbidityHumoral Hypercalcemia of Malignancy (for patients who do not respond adequately to bisphosphonates)Prevent bone-related complications in multiple myeloma
