Mechanism
Levodopa, a metabolic precursor of dopamine, is decarboxylated to dopamine, which replaces the deficient neurotransmitter in Parkinson's disease. Carbidopa, a dopa-decarboxylase inhibitor, prevents the peripheral decarboxylation of levodopa, increasing its availability to the brain. Entacapone, a catechol-O-methyltransferase (COMT) inhibitor, further reduces the peripheral metabolism of levodopa, thus prolonging its therapeutic effect.
Indications
Parkinson’s disease and end-of-dose motor fluctuations not adequately controlled with levodopa and dopadecarboxylase inhibitor treatment
Dosing
- Adult
- 1 tablet per dose. Maximum daily dose varies by formulation: STALEVO 100/25/200, 125/31.25/200, 150/37.5/200, 50/12.5/200, 75/18.75/200: maximum 10 tablets per day. STALEVO 175/43.75/200: maximum 8 tablets per day. STALEVO 200/50/200: maximum 7 tablets per day.
