Mechanism

Barbiturates enhance GABA-mediated inhibitory transmission throughout the CNS. They bind to an allosteric site on the GABAA receptor, increasing the mean open time of the GABA-activated Cl– channel. At higher concentrations, barbiturates directly activate channel opening.
