Barbiturates act primarily at the GABAA: BZD receptor–Cl¯ channel complex and potentiate GABAergic inhibition by increasing the lifetime of Cl¯ channel opening induced by GABA. At high concentrations, they directly increase Cl¯ conductance (GABA-mimetic action) and inhibit Ca2+ dependent release of neurotransmitters. They also depress glutamate induced neuronal depolarization through AMPA receptors.
