Mechanism

Ozogamicin is the cytotoxic component of antibody-drug conjugates. The antibody component (e.g., gemtuzumab or inotuzumab) targets specific tumor antigens (e.g., CD33 or CD22) and facilitates internalization into tumour cells. Once inside, ozogamicin (a calicheamicin derivative) is cleaved and causes double-strand breaks in DNA, leading to cell cycle arrest and apoptotic cell death.
