Mechanism

A recombinant human monoclonal anti-FGF23 antibody that binds to FGF23 and inhibits its activity, leading to increased phosphate reabsorption and increased serum concentrations of phosphate and calcitriol.

Indications

X-linked hypophosphatemia (XLH)FGF23-related hypophosphatemia in tumor-induced osteomalaciaOsteogenic osteomalacia (to improve metabolic features and potentially bone structure and fracture risk)

Dosing

Adult
0.5 mg/kg every 4 weeks (can be increased to 2 mg/kg every 2 weeks)
Pediatric
1 mg/kg every 4 weeks for X-linked hypophosphatemia.
Renal adjustment
Contraindicated in severe kidney impairment or end-stage kidney disease.

Contraindications

  • oral phosphate and active vitamin D analogues
  • serum phosphorus within or above the normal limits
  • severe kidney impairment
  • end-stage kidney disease
