Mechanism

PDE-3 inhibitor which increases intracellular cAMP in platelets and vascular smooth muscle resulting in antiaggregatory and vasodilator effects.

Indications

Intermittent claudication (in patients with no rest pain or heart failure)

Dosing

Adult
100 mg BD, 30 min before or 2 hour after food

Half-life
approximately 12 hours
Metabolism
extensively metabolized by CYP3A4 and CYP2C19 into active and inactive metabolites
Excretion
in urine

Contraindications

  • patients with heart failure
  • patients who have pain even at rest
  • patients with tissue necrosis
  • inhibitors of CYP3A4
  • inhibitors of CYP2C19
