Mechanism
DFP produces virtually irreversible inactivation of acetylcholinesterase (AChE) and other esterases by alkylphosphorylation of the active center serine. The stability of the phosphorylated enzyme is enhanced through 'aging,' which results from the loss of one alkyl group, making it resistant to reactivation.

Bioavailability
High lipid solubility, low molecular weight, and volatility facilitate inhalation toxicity, transdermal absorption, and penetration into the CNS.
