Mechanism

Rotigotine preferentially binds to D2 and D3 receptors and has much lower affinity for D1 receptors. Additionally, rotigotine acts as an agonist at 5-HT1A and 5-HT2 receptors and an antagonist at α2 adrenergic receptors.

Treatment is usually started with the 2 mg/24 hours patch and titrated weekly as necessary by increasing the patch size to 4 mg/24 hours and then to 6 mg/24 hours.
