Mechanism

Chlorothiazide inhibits NaCl transport in the distal convoluted tubule by blocking the Na+-Cl− symporter (ENCC1 or TSC) on the luminal membrane of DCT epithelial cells. Some thiazide diuretics also weakly inhibit carbonic anhydrase.
