Dibucaine acts at the cell membrane to prevent the generation and the conduction of nerve impulses. It blocks nerve impulse conduction by directly interacting with voltage-gated Na+ channels, decreasing or preventing the transient increase in Na+ permeability that occurs with membrane depolarization. This action increases the threshold for electrical excitability and slows impulse conduction, ultimately leading to a failure of nerve propagation.
