Adapalene, a third-generation retinoid, selectively binds to nuclear retinoic acid receptors (RARs), particularly RAR-gamma. This binding activates specific DNA sequences (RAREs), modulating gene expression involved in cellular differentiation and keratinization of follicular epithelial cells. The resulting normalization of follicular keratinization, decreased keratin and sebum production, and anti-inflammatory effects suppress comedone formation and reduce inflammatory lesions in acne.
Contraindicated in women who are pregnant or planning pregnancy; women of childbearing potential must use effective contraception (progestogen-only contraceptives not considered effective).
